Dedicated in 1855 as the uptown (25th Street was uptown at that point) chapel of Trinity Church, the structure was designed by the same architect as the mother church, Richard Upjohn. The brownstone edifice was the venue of the1885 marriage of Edith Jones and Edward Wharton. Edith Wharton later used the church as a setting in her novel 'Age of Innocence." The church suffered in the early 20th century from the same action that had created it, the movement of parishioners uptown.

In 1942, the Episcopal Diocese sold the complex to the growing Serbian community. Rechristened the Cathedral of St. Sava, it became the first Serbian Orthodox church on the east coast. The later 20th century saw some of the Protestant ornamentation replaced with Byzantine-style stained glass and Serbia iconography. On the evening of May 1, 2016, the day of Orthodox Easter celebrations, the church suffered a devastating fire. While the roof and most of the interior was completely destroyed, the stone walls of the church remained. As of December of 2017 the work was being completed on the church to clear out some of the damaged interior and erect scaffolding for the eventual reconstruction of the building.
